Experimental Flowpath

We have worked with several industry partners, and have optimized our service flowpath.

Step 1: Meeting to determine your goals, and signing contracts

We want to ensure your project is amenable to HDX-MS. Let us know your goals, and if it fits with our platform we can sign contracts and move to the next step. 

 

Step 2: Generation of a high quality non-deuterated peptide map 

Before setting a full HDX experiment, we recommend validation experiments to ensure that the project is amenable to HDX-MS

The goal of this experiment is to:

  1. Optimize peptide cleavage conditions

  2. Determine optimal protein concentrations

  3. Obtain a peptide map and ensure that all regions of interest are covered

Step 3: Hydrogen Deuterium eXchange (HDX-MS) 

After discussing the goals of the experiment with you, we will determine the number of conditions to test. Each condition tested will include 3-4 timepoints of deuterium incorporation in triplicate to determine if there are any significant differences in deuterium incorporation between conditions. For a given condition this will require 16-20 samples (experiments set in quadruplicate to ensure high quality triplicate data). MS data will be analyzed in HDExaminer, with the output being the percent deuterium incorporation at each timepoint. We will meet with you to ensure the data is correctly interpreted.

HDX-MS data will be delivered no later than two months after delivery of samples, with a goal of four weeks
